Morphological Study of the Renal Juxtaglomerular Granular Cells - Effects of DCA on the Adrenalectomized Mice

Abstract
The juxtaglomerular granular cells of the male white mice were investigated by light and electron-microscopies with the aim elucidating the morphological characteristics of the graunles and juxtaglomerular granular index (JGI) and granular cell index (GCI) were calculated under the various experimental conditions. The experimental groups were classified as the normal control, one-, two, and four-week-survived after adrenalectomy, and some of the each group were administered DCA in dose of 1§·/§¸ body weight. For the light microscopical study, after the fixation in Zenker-formol solution the sections were stained by Wilson¡¯s method. And for the ultrastructural study, under the anesthesia of nembutal the kidney was fixed by purfusion method with 2.3% glutaraldehyde in 45mM cacodylate buffer following prerinzing with heparinated 0.1% Ringer-lidocaine solution. After the removal of the tissue, it was postfixed in 0.1% osmium tetroxide in 45mM cacodylate buffer by immersion, and the thin sections were stained with uranyl acetate and lead citrate. The results observed were as follows; 1. Adrenalectomy increased both JGI and GCI markedly, especially in 2-weeks¡©survived group after adrenalectomy. 2. Increment of JGI was chiefly due to the increase of Type 3 granular cells. 3. There wasn¡¯t any significant decrease in both GCI and JGI following administration of DCA to normal animals. 4. By the administration of DCA during the survival period after adrenalectomy, JGI and GCI showed slight decrease but not expected to return to normal value. 5. In the juxtaglomerular granular cells in normal control, well-developed rough surfaced endoplasmic reticulum and round secretory grarnules were found. 6. In the adrenalectomized group, secretory granules showed amorphous and coalesced large granules, and the prototype of secretory granules are found near the Golgi apparatus. 7. In one of the juxtaglomerular cells secretory glanules not surrounded with single membrane are found within the cisternae of rough surfaced endoplasmic reticulum. In the other cell, markedly developed rough surfaced endoplasmic reticulum filled whole cytoplasm. In this case, there were no secretory granules and only small-sized mitochondria were found between them. 8. It is difficult to discuss that the administration of DCA causes ultrastructural changes in juxtaglomerular granular cells.
